A complete guide to choosing, hydrating, pasteurising and sterilising substrates for gourmet mushroom cultivation.
Choosing and preparing the correct substrate is one of the most important parts of mushroom cultivation. The substrate provides the moisture, nutrition and physical structure that mushroom mycelium needs while it colonises and prepares to produce mushrooms.
The correct preparation method depends on the material. Relatively low-nutrient substrates such as clean straw are commonly pasteurised, while grain and hardwood supplemented with wheat bran normally require pressure sterilisation and clean inoculation.

Getting startedWhat Is Mushroom Substrate?
Mushroom substrate is the organic material through which mushroom mycelium grows. Unlike plants, fungi do not use photosynthesis. Mycelium releases enzymes into a suitable food source, breaks down the material and absorbs the nutrients needed to establish a colony.
Nutrition
The material must provide a suitable food source for the selected mushroom species.
Moisture
The substrate needs enough available water for growth without becoming waterlogged.
Structure
Air spaces help prevent dense, wet areas and support healthy colonisation.
Clean preparation
Pasteurisation or sterilisation reduces competition from moulds and bacteria.
Common mushroom substrates include hardwood sawdust, hardwood pellets, wheat straw, hardwood woodchips, sterilised grain, prepared compost, coco coir-based mixtures and suitable hardwood logs.
Coffee grounds and cardboard can support some mushroom growth, but they are less reliable as stand-alone beginner substrates. Coffee grounds are nutritious, compact easily and can contaminate rapidly after brewing.

Preparation methodPasteurisation vs Sterilisation
Pasteurisation and sterilisation are different processes. The correct method depends mainly on the nutritional content of the substrate.
Pasteurisation
Pasteurisation reduces competing organisms without attempting to eliminate every microorganism. It is commonly used for relatively low-nutrient materials such as clean straw and some outdoor woodchip methods.

- Wheat, barley or oat straw
- Some unsupplemented sawdust methods
- Hardwood woodchips used outdoors
- Other low-nutrient bulk materials
Sterilisation
Sterilisation uses higher heat under pressure to prepare a much cleaner growing medium. It is normally required for materials rich enough to support rapid mould and bacterial growth.
Common uses
- Wheat, rye, millet and other grain
- Hardwood supplemented with wheat bran
- Agar media
- Liquid culture media
Substrate method 1How to Pasteurise Straw for Oyster Mushrooms
Use clean, dry wheat, barley or oat straw that is free from visible mould, chemical treatment and unpleasant odours.

Equipment Needed
- Clean wheat, barley or oat straw
- A large heat-resistant container
- Hot water
- A reliable thermometer
- A clean mesh bag, basket or pillowcase
- A clean draining and cooling area
- Fully colonised Oyster mushroom grain spawn
- A suitable grow bag or bucket

Chop the straw
Cut the straw into pieces approximately 3 to 8 cm long. Shorter pieces are easier to hydrate, mix and pack, and create more contact points through which the mycelium can spread.

Hydrate the straw
Very dry straw can be soaked before heating so it absorbs water more evenly. An overnight soak is optional rather than essential.

Heat the water
Heat enough water to submerge the straw. Aim for approximately 65°C to 75°C. Avoid a vigorous boil, which can make the straw overly soft and difficult to drain.

Pasteurise
Submerge the straw and maintain the target temperature for approximately 60 to 90 minutes. Use a clean heat-resistant weight if necessary to keep the material below the water.

Drain and cool
Remove the straw and allow it to drain thoroughly. Leave it in the mesh bag or spread it across a clean surface until it has cooled to room temperature.

Check the moisture
Squeeze a representative handful firmly. The straw should feel thoroughly damp, but only a few drops should appear. If water streams from it, allow it to drain for longer.

Add grain spawn
Mix clean, fully colonised Oyster mushroom grain spawn evenly through the cooled straw. Several evenly distributed inoculation points are more useful than one thick layer of spawn.

Pack the container
Pack the inoculated straw into a clean mushroom grow bag or bucket. Fill it firmly enough to remove large gaps, but do not compress it into a dense, airless mass.
Substrate method 2How to Prepare Supplemented Hardwood Substrate
Supplemented hardwood is widely used for Lion’s Mane, Shiitake, Chestnut, Maitake, Reishi and many Oyster mushroom varieties. Hardwood provides the main structure and carbon-rich food source, while wheat bran adds readily available nutrition.

- 85% clean hardwood sawdust or hardwood pellets
- 15% wheat bran
- Water added gradually until field capacity is reached
This is a useful starting point rather than a universal formula. Ingredient density and water absorption vary between brands and batches.

Choose clean hardwood
Oak and beech are commonly used. Avoid pressure-treated, painted, stained or varnished wood, MDF, chipboard, plywood, glued timber, oily workshop sawdust and strongly resinous softwoods.

Mix the dry ingredients
Combine the hardwood and wheat bran thoroughly before adding water. Even distribution prevents concentrated pockets of supplement.

Add water gradually
Add most of the calculated water first, mix thoroughly and allow hardwood pellets time to soften and expand. Add the remaining water in small amounts.

Test field capacity
Take a handful from the middle of the batch and squeeze it firmly. It should hold together briefly and release no more than a small number of drops.

Fill filter-patch grow bags
Remove large air pockets, keep the filter patch clean and avoid packing the material into an extremely dense block.

Pressure sterilise
Use an appropriate sterilisation cycle for the bag size, density and equipment. Large, tightly packed substrate bags take longer for the centre to reach sterilisation temperature than small jars.

Cool completely
Allow the equipment to depressurise naturally and keep the bags sealed while they cool. Do not inoculate until the centre of the substrate has returned completely to room temperature.
Moisture controlHow to Test Mushroom Substrate Field Capacity
Field capacity is the moisture level at which substrate contains enough water for mycelial growth without becoming waterlogged.

Correct
- The material holds together briefly.
- A small number of drops may appear.
- It breaks apart again when disturbed.
Too wet
- Water streams from the material.
- The mixture feels muddy or heavily compacted.
- Water collects in the bottom of the bag.
Too dry
- The material remains dusty or loose.
- It will not hold together when squeezed.
- Dry pellets or pale patches remain.
Test several parts of a large batch because moisture may not be evenly distributed.

Liquid culture
Liquid culture is living mushroom mycelium suspended in sterile nutrient liquid. It is normally used to inoculate sterilised grain or a compatible all-in-one grow bag through a self-healing injection port.
Grain spawn
Grain spawn is sterilised grain that has already been colonised by mushroom mycelium. It is ready to mix through a compatible fruiting substrate such as pasteurised straw or sterilised hardwood.
Standard bulk-substrate workflow
- Inoculate sterilised grain with a clean liquid or agar culture.
- Allow the grain to colonise completely.
- Break the colonised grain into separate kernels.
- Mix the grain spawn evenly through the prepared bulk substrate.
- Allow the new substrate to colonise.
- Introduce the fruiting conditions required by the species.

- Do not open the bag.
- Do not add water or wheat bran.
- Do not pasteurise or sterilise it again.

- Clean the black injection port and allow the alcohol to evaporate.
- Inject the liquid culture into the lower grain layer.
- Keep the bag sealed while the grain begins to colonise.

ColonisationConditions and Signs of Healthy Growth
Keep inoculated bags or containers in a clean area away from direct sunlight, radiators, damp floors and strong draughts. Temperature requirements vary by mushroom species, so check the needs of the selected culture.
Complete darkness is not normally essential. Stable temperature, correct moisture, healthy culture, suitable gas exchange and clean preparation are more important during colonisation.

Using the wrong substrate
Match the food source to the mushroom species rather than using one recipe for every culture.
Pasteurising a nutrient-rich mixture
Grain and hardwood supplemented with wheat bran normally require pressure sterilisation.
Adding too much water
Waterlogged substrate compacts, reduces air spaces and favours bacterial growth.
Leaving the substrate too dry
Mycelium requires available water. Dry pockets can slow or stall colonisation.
Inoculating while warm
The outside of a large bag may feel cool while its dense centre remains hot.
Using liquid culture in ordinary bulk substrate
Liquid culture is not a substitute for evenly distributed grain spawn unless the bag is specifically designed as an all-in-one injection bag.
Blocking the filter patch
Keep the patch clean, dry and unobstructed so the mycelium can exchange gases.
Relying on old coffee grounds
Coffee grounds can contaminate quickly and compact into wet, poorly aerated masses.
Frequently asked questionsMushroom Substrate FAQ
Can liquid culture be injected directly into mushroom substrate?
Only when the receiving material is sterile and designed for direct injection, such as sterilised grain or a compatible all-in-one grow bag with a self-healing injection port.
For ordinary straw, hardwood bulk substrate, woodchips or CVG, first produce clean, fully colonised grain spawn and then mix the spawn through the prepared substrate.
Can supplemented hardwood be pasteurised?
Supplemented hardwood is nutrient-rich and should normally be pressure sterilised. Pasteurisation does not provide the same clean starting point.
How wet should mushroom substrate be?
It should be at field capacity. When squeezed firmly, it should hold together briefly and release no more than a small number of drops. It should not stream water.
Can I use softwood pellets?
Hardwood pellets are the safer general choice for common wood-loving gourmet mushrooms. Use strongly resinous softwood only when a proven species-specific method calls for it.
Can I use fresh coffee grounds?
Fresh coffee grounds can support Oyster mycelium, but they contaminate and compact easily. Use them promptly, keep the proportion modest and combine them with a more structured material.
How long can prepared substrate be stored?
Pasteurised substrate is best inoculated promptly after cooling and draining. A sealed sterilised bag can remain clean for longer, but freshness, seals, moisture and storage conditions all matter.
Do I need to add water to an Artisan Mushrooms grow bag?
No. The bag is supplied correctly hydrated and pressure sterilised. Opening it or adding water increases contamination risk.
Should I mix an all-in-one grow bag immediately?
No. Allow visible mycelium to establish through the lower grain layer before mixing it through the hardwood at the stage recommended in the product instructions.

Successful substrate preparation comes down to five decisions: match the substrate to the mushroom, hydrate it correctly, choose pasteurisation or sterilisation according to its nutritional content, allow complete cooling and use the correct inoculum for the growing system.
