How to Inoculate grow bags, grain and substrates
A complete beginner-friendly guide to clean mushroom inoculation, grain and grow-bag colonisation, break and shake, healthy mycelium and contamination warning signs.
Inoculation introduces a clean mushroom culture into a compatible sterile growing medium. Colonisation is the period in which the mycelium establishes itself and spreads through the available grain or substrate.
These stages are often described together, but the correct method depends on what you are inoculating. Liquid culture is commonly used for sterilised grain or a compatible all-in-one grow bag. Fully colonised grain spawn is used for ordinary bulk substrate.
- Use liquid culture to inoculate sterilised grain or a compatible all-in-one grow bag through a self-healing injection port.
- Use fully colonised grain spawn to inoculate ordinary hardwood, straw or CVG bulk substrate.
- Use a new sterile needle, wipe the injection port and allow the alcohol to evaporate before inserting the needle.
- Do not open a sealed injection-port bag, add water or cover its filter patch.
- Do not mix an all-in-one bag immediately after injection. Allow the grain layer to establish first.
- Colonisation time and mycelium appearance vary considerably between mushroom species.
Start here Choose the Correct Inoculation Route
The word substrate is sometimes used loosely, which can make beginner instructions confusing. Sterile grain, an all-in-one grow bag and ordinary bulk substrate are not used in exactly the same way.
Liquid culture → sterilised grain
Inject clean liquid culture into sterilised wheat, rye, millet or another suitable grain. Once completely colonised, the grain is called grain spawn.
Liquid culture → compatible all-in-one bag
A purpose-designed all-in-one bag can be inoculated directly when it contains a suitable sterile grain layer and a self-healing injection port.
Grain spawn → bulk substrate
Fully colonised grain spawn is mixed evenly through compatible hardwood, straw, CVG or another species-appropriate fruiting substrate.
Agar → sterilised grain
Clean agar wedges can be transferred to sterilised grain inside a still-air box or in front of a properly operated laminar-flow hood.
Before inoculation Prepare a Clean, Safe Workspace
Injection-port inoculation is a closed process and does not require opening the bag. It is more forgiving than agar or grain-to-grain transfer, but careful handling still matters.
Prepare the area
- Choose a clean table away from open windows, fans and strong draughts.
- Remove pets, food, potting compost and unnecessary items.
- Wash and dry your hands thoroughly.
- Clean the work surface and allow it to dry.
- Wear clean gloves when practical.
- Keep the sterile needle packaged until immediately before use.
Check the receiving bag
- Confirm that the plastic, seams, filter patch and injection port are intact.
- Do not use a bag that is punctured, leaking or visibly contaminated.
- Keep the bag sealed and avoid pressing substrate tightly against the filter patch.
- Allow a refrigerated culture and a very cold bag to move closer to normal working temperature while remaining sealed.
Beginner method How to Inoculate an All-in-One Hardwood Grow Bag
The Artisan Mushrooms ready-to-inoculate hardwood grow bag contains a lower grain layer, hardwood substrate, a filter patch and a black self-healing injection port. It arrives hydrated, pressure sterilised and sealed.

Inspect the sealed bag
Check the plastic, seals, filter patch and black injection port. Do not open the bag, add water or sterilise it again.

Distribute the liquid-culture mycelium
Shake the sealed syringe vigorously so that visible mycelium is broken into smaller pieces and distributed through the liquid.

Attach a new sterile needle
Hold the needle by its plastic hub. Keep the protective cap fitted until immediately before inoculation and do not touch the metal needle.

Clean the injection port
Wipe the black port with the supplied alcohol wipe and allow it to air-dry completely.

Inject the lower grain layer
Insert the needle through the centre of the black port and direct the liquid into the grain layer at the bottom. For an Artisan Mushrooms 2 kg bag, approximately 5 to 10 ml is generally suitable.

Withdraw the needle carefully
Remove it without twisting or puncturing the clear bag plastic. The port reseals around the puncture. It does not normally require tape.

Label the bag
Record the mushroom species, culture, inoculation date and any useful batch information.

Leave the bag sealed
Place it in a clean location with the filter patch dry and unobstructed. Do not mix the bag immediately after inoculation.
Producing grain spawn How to Inoculate a Sterilised Wheat Grain Bag
A sterilised wheat grain bag contains prepared grain but no mushroom culture. After inoculation and complete colonisation it becomes grain spawn, which can then be mixed through a compatible fruiting substrate.

Check the bag and grain
The bag should remain sealed, with intact plastic, a clean filter patch and no slimy, discoloured or foul-smelling grain.

Shake the liquid culture
Break up and distribute the mycelium throughout the syringe before attaching the sterile needle.

Wipe and dry the port
Clean the self-healing injection port and wait for the alcohol to evaporate.

Inject the grain
Insert through the port and distribute small amounts across several nearby areas. Approximately 5 to 10 ml is generally suitable for an Artisan Mushrooms 2 kg wheat grain bag.

Label and incubate
Keep the bag sealed, upright and spaced from other colonising bags so heat can escape.
Intermediate methods Agar-to-Grain and Grain-to-Grain Transfers
Agar and grain-to-grain transfers require opening the receiving container. They should be carried out inside a clean still-air box or in front of a correctly operated laminar-flow hood.
Agar to grain
Cut several small wedges from clean, healthy mycelium and transfer them into sterilised grain using a sterile scalpel. Minimise the time that both the plate and receiving bag remain open.
Grain to grain
Transfer a small quantity of clean, fully colonised grain into a fresh sterilised grain bag. Each colonised kernel becomes a new growth point.
- Clean the outside of receiving containers before placing them in the work area.
- Work deliberately rather than moving quickly and creating turbulence.
- Keep hands and tools downstream of clean laminar airflow.
- Do not move unclean items over open plates, jars or bags.
- Close and seal receiving containers immediately after transfer.
Incubation Suitable Colonisation Conditions
There is no single perfect colonisation temperature or timeline for every mushroom. Species, strain, bag size, inoculation method and culture vitality all affect the spawn run.
Temperature
A practical general range for many gourmet species is approximately 20°C to 25°C. Always check the selected culture because some species prefer cooler or warmer conditions.
Light
Complete darkness is not normally required. Low ambient light is acceptable, but avoid direct sunlight and strong heat-producing lights.
Gas exchange
Keep the filter patch clean, dry and unobstructed. Do not cover it with tape or compress it tightly against the grain or substrate.
Room humidity
A sealed bag retains its own substrate moisture. High external room humidity is not normally required during colonisation.

Growth distribution When to Break, Shake or Mix the Bag
Loose grain bag
Once a clearly established section of healthy mycelium is visible, a grain bag can often be broken up and shaken once to distribute the colonised kernels. This is commonly done at roughly one-quarter to one-third colonisation, unless the product or species instructions say otherwise.
Artisan Mushrooms all-in-one bag
Do not massage the bag immediately after liquid-culture inoculation. Allow the mycelium to establish strongly through the lower grain layer. At the appropriate stage, break up the colonised grain and mix it through the hardwood substrate.
After mixing, gently tamp the bag down to reform a reasonably compact block. Trying to press it into an extremely hard block is unnecessary and can remove useful air spaces.

A properly mixed hardwood block is normally left to colonise and consolidate. Repeated aggressive handling can damage the filter, puncture the bag, compact the substrate and interrupt developing mycelial connections.
Monitoring growth Healthy Mycelium and Contamination Warning Signs
Common signs of healthy growth
- White or cream-coloured mycelium
- Gradual expansion from inoculation points
- Fine, cotton-like, feathered or rope-like growth
- Grain becoming connected and held together
- Species-appropriate consolidation and maturation
Possible contamination signs
- Green, black, pink or red growth
- Wet, greasy, slimy or burst grain
- Rapidly expanding growth unlike the expected mycelium
- Large stagnant wet areas
- Sour, rotten or fermented odours
- Damaged plastic, filter patch or seals
Species differences that can look unusual
- Lion’s Mane: often fine, wispy and less densely white than Oyster mycelium.
- Shiitake: may form raised white growth and later develop normal brown colouring during maturation.
- Reishi: can become thick and white and may produce amber-coloured droplets.
- Older healthy mycelium: can produce small quantities of yellow or amber metabolites, especially under stress.
Problem solving Troubleshooting Slow or Stalled Colonisation
No visible growth yet
Some cultures recover slowly after inoculation. Visible growth can take several days or one to two weeks, particularly with slower species or a direct-inoculated hardwood bag.
Growth started and stopped
Check temperature stability, filter-patch obstruction, excessive wetness, bag damage and whether the culture was healthy and stored correctly.
Only one area is colonising
This can be normal early in the spawn run. A grain bag may later benefit from one break and shake. Do not repeatedly disturb an all-in-one bag before the grain has established.
Heavy condensation
Some condensation can result from temperature differences and mycelial heat. Persistent pooling, slimy grain or a sour smell is more concerning than clean condensation alone.
The bag feels very warm
Space the bags apart, move them away from radiators and check the surrounding temperature. Do not place colonising bags in a hot press or sealed insulated box.
The filter patch is wet or blocked
Reposition the sealed bag so the patch is exposed and can dry. Do not cut, puncture, tape over or inject through the filter.
Completing colonisation How to Know When the Bag Is Ready
Sterilised grain bag
Grain spawn is ready when the intended grain is evenly colonised, individual kernels are connected by healthy mycelium and no large uncolonised, wet or discoloured areas remain.
Hardwood all-in-one or fruiting block
The block should show species-appropriate mycelium throughout, feel firm and hold together. Some species benefit from additional consolidation after visible colonisation.
- Oyster: often moves to fruiting relatively soon after full colonisation.
- Lion’s Mane: can begin fruiting early inside a bag, so follow the species-specific instructions.
- Shiitake: normally requires a longer maturation period and brown outer surface.
- Maitake and Reishi: can require additional consolidation before fruiting.
When the block is ready, follow the correct fruiting method for the mushroom species. Cutting an X, removing the top of a bag, applying a casing layer or moving a block into a fruiting chamber are not interchangeable instructions.
Frequently asked questions Mushroom Inoculation and Colonisation FAQ
Can liquid culture be injected directly into mushroom substrate?
Only when the receiving material is sterile and specifically designed for direct inoculation, such as sterilised grain or a compatible all-in-one bag with a grain layer and self-healing injection port. Ordinary bulk substrate should normally be inoculated with fully colonised grain spawn.
How much liquid culture should I use in a 2 kg bag?
Approximately 5 to 10 ml is generally suitable for Artisan Mushrooms 2 kg sterilised grain and all-in-one hardwood bags. Follow the product instructions if using another container. Excess liquid is not helpful.
Should I flame-sterilise a newly opened sterile needle?
No. A fresh packaged needle is already sterile and does not normally require flame sterilisation before its first use. Never use a flame inside a still-air box or near alcohol vapour.
Should I inject through the filter patch?
No. Inject only through the black self-healing port. The filter patch must remain intact, clean, dry and unobstructed.
Should I tape over the injection port afterwards?
A correctly fitted self-healing port normally reseals around the needle puncture and does not require tape.
Do colonising bags need complete darkness?
No. Low ambient room light is normally acceptable. Avoid direct sunlight because it can overheat the bag and cause large temperature changes.
Should I mix the bag immediately after inoculation?
No. Allow the culture to recover and establish. A loose grain bag can later be broken and shaken once. In an Artisan all-in-one bag, allow the lower grain layer to colonise strongly before mixing it through the hardwood.
Can I open the bag to smell it?
No. Keep a suspicious bag sealed. Sour or rotten odours may sometimes be detectable through the filter patch, but the bag should not be opened simply to check the smell.
Why is Lion’s Mane mycelium difficult to see?
Lion’s Mane often produces finer, wispy and less densely white mycelium than Oyster mushrooms. Judge it according to the species, growth progression and absence of contamination signs.
How long should colonisation take?
It depends on the mushroom, culture, bag size, temperature and inoculation method. Visible growth may appear within several days or take one to two weeks. A grain bag may require several weeks, while a directly inoculated all-in-one hardwood bag can require four to twelve weeks or longer for slower species.
